I see two bands on my western blot. One is 50kda, and one is 20kda. Is this normal? | |
The 20kda is the protein of interest, the histone H2A.X. This antibody usually produce a 50kd non-specific band in western blot, but this do not affect its detection and specificity.

Y011268 is a phospho-antibody and the signal strength is dependent on how many of the H2A.X proteins are phosphorylated at the Serine 139 site. Many methods can stimulate the phosphorylation of this site, such as UV and ionizing radiation. Some methods that can stimulate MAPK / JNK pathway can also stimulate this site. It is important to maintain a strong level of phosphorlyation for the protein when producing cell lysate. We suggest optimizing the stimulating method for the sample to increase the detection from the antibody.
